The PAM is about 2-6 nucleotides downstream of the DNA sequence targeted by the guide RNA and the Cas nuclease cuts 3-4 nucleotides upstream of it. PAM sequences. The canonical PAM is the sequence 5'-NGG-3', where "N" is any nucleobase followed by two guanine ("G") nucleobases.
